He was born to a golf enthusiastic family und began playing Golf in the Circolo Golf Torino. In the peak of playing amateur Molinari won as first continental European the US Amateur Championship.
He turned professional in 2006 and went on the Challenge Tour. There he won his first tournament in 2007. After three wins more in 2009 Molinari automatically qualified for the European Tour. In that year Molinari and his younger brother Francesco became the first Italians winning the World Cup
In all Molinari won twice on the European Tour so far and was consequently picked to play the Ryder Cup Matches by Colin Montgomerie.
